Delivery System
The ability of Catalase-SKL
to gain entry into the peroxisome and, once there, to metabolize hydrogen
peroxide, is dependent on the enzyme first having accessed the cell interior.
That is, catalase must be delivered inside the cell. To achieve this goal, EXT
has adopted a protein “transduction” strategy, whereby Catalase-SKL is either
mixed with a delivery molecule prior to the addition to cells, or a delivery
motif is fused to the end of the catalase molecule (opposite of where the SKL
signal is positioned). By either method, Catalase-SKL is transduced across the
cell’s plasma membrane and enters the cytosol. Catalase-SKL is then poised to
engage the peroxisomal protein import apparatus and enter the peroxisome.
Experiments are currently underway to optimize Catalase-SKL delivery
efficiencies for a wide range of cell and tissue types.
